Correlative Cellular Mass Spectrometry Imaging and Amperometry Show Dose Dependent Changes in Lipid Composition and Exocytosis

Abstract Aberrant functioning of the proteasome has been associated with crucial pathologic conditions including neurodegeneration. Yet, complex underlying causes at cellular level remain unclear and there are conflicting reports neuroprotective to neurodegenerative effects proteasomal inhibitors such as lactacystin that utilised models for diseases. The results may be different dose regimes hence we have performed a dependent study identify concurrent changes in cell membrane lipid profile dynamics exocytosis using combination surface sensitive mass spectrometry single amperometry. Significant negatively charged lipids were doses showed weak correlation while PE PE−O correlated initial pore formation total release vesicle content respectively.
